چکیده

Hemophilia A is caused by a deficiency of actor-Vlll procoagulant (NIII) activity. The current treatment by frequent infusions of plasma-derived Nlll concentrates is very effective but has the risk of transmittance of blood-borne viruses (human immunodeficiency virus IHIVI, hepatitis viruses). Use of recombinant DNA-derived Nlll as well as gene therapy could make hemophilia treatment independent of blood-derived products. So far, the problematic production of the Nlll protein and the low titers of the Nlll retrovirus stocks have prevented preclinical trials of gene therapy for hemophilia A in large-animal models. We have initiated a study of the mechanisms that oppose efficient Nlll synthesis. We have established that Nlll cDNA contains sequences that

Bacterial Expression and Functional Characterization of A Naturally Occurring Exon6-less Preprochymosin cDNA

Chymosin (Rennin EC 3.4.23.4), an aspartyl proteinase, is the major proteolytic enzyme in the fourthstomach of the unweaned calf, and it is formed by proteolytic activation of its zymogene, prochymosin.Following the cloning of synthesized cDNAs on mRNA pools extracted from the mucosa of the calf fourthstomach, we have identified an alternatively spliced form of preprochymosin ...

The human clotting factor VIII cDNA contains an autonomously replicating sequence consensus- and matrix attachment region-like sequence that binds a nuclear factor, represses heterologous gene expression, and mediates the transcriptional effects of sodium butyrate.

Expression of the human blood-clotting factor VIII (FVIII) cDNA is hampered by the presence of sequences located in the coding region that repress transcription. We have previously identified a 305-bp fragment within the FVIII cDNA that is involved in the repression (R.C. Hoeben, F.J. Fallaux, S.J. Cramer, D.J.M. van den Wollenberg, H. van Ormondt, E. Molecular Study of Vascular Endothelial Growth Factor Gene in Iranian Patients after Myocardial Infarction

Background: Stimulation of collateral artery growth (arteriogenesis) and/or capillary network growth (angiogenesis) would be beneficial to the patients with myocardial infarction. To understand the central role of vascular endothelial growth factor (VEGF) in biological angiogenesis, we performed molecular analysis of the VEGF gene in patients afflicted with acute myocardial infarction (AMI). Me...

Cloning and Characterization of cbhII Gene fromTrichoderma parceramosum and Its Expressionin Pichia pastoris

The genomic and cDNA clones encoding cellobiohydrolase II (CBHII) have been isolated and sequenced from a native Iranian isolate of Trichoderma parceramosum, a high cellulolytic enzymes producer isolate. This represents the first report of cbhII gene from this organism. Comparison of genomic and cDNA sequences indicates this gene contains three short introns and also an open reading frame codin...
